The short version: NMN and NR really do raise NAD+ in your blood — that part is true. But there is no solid proof yet that they slow ageing, add years, or do the dramatic things you have seen in headlines. Most of that comes from mice, not people. Here is the honest picture. First, what is NAD+?
NAD+ is a helper molecule your cells use to turn food into energy and to repair themselves. Think of it as a tiny rechargeable battery that keeps your cells running.
Here is the catch: NAD+ slowly drops as you get older. So the idea sounds simple — top it back up and maybe you slow ageing. That is the hope behind the whole industry. It is a reasonable idea. It just has not been proven.
You cannot swallow NAD+ directly — your gut breaks it down before it is useful. So supplements use “precursors”, which are building blocks your body turns into NAD+. The two popular ones are NMN and NR.

What does the science actually show in people?
Human studies exist, but most are small, short, and measure lab markers rather than real-life results. Here is the honest read.
NAD+ in your blood rises — reliably. This is the most consistent finding. Across trials, blood NAD+ climbed by roughly three-quarters at a 500 mg daily dose of NMN, and more than doubled at 1,000 mg. NR shows the same upward move. So yes, the “raises NAD+” promise holds up.
Real benefits are hit-or-miss. The most cited win is a 2021 study in the journal Science: NMN at 250 mg a day improved how well muscle responded to insulin — but only in one specific group, prediabetic women past menopause. Meanwhile, several trials of NR in older and overweight men found no improvement in insulin response at all. A 2024 review that pooled nine trials found only small, modest effects on blood-sugar markers and muscle strength.
No proof of a longer life. No study has followed enough people for long enough to say these add healthy years. Anyone who promises that is selling, not citing.
The bottom line of the research: these molecules clearly change your blood chemistry. They have not been shown to change how you actually age.
So where does all the hype come from?
A lot of it traces back to one charismatic Harvard scientist, David Sinclair, whose book and constant media presence did more to sell the NAD+ dream than any single study. The problem is that he is not a neutral referee. He co-founded companies with money riding on these molecules — including Metro Biotech, whose drug paperwork on NMN briefly got it pulled from US shelves (more on that below).
In 2024, Sinclair stepped down from leading a major longevity research group after fellow scientists pushed back hard on his “age reversal” claims. Respected researchers, such as biochemist Charles Brenner, have been openly critical. The lesson: a great story plus a financial interest is not the same thing as proof.
Why the mouse results do not carry over
Most of the exciting headlines come from mice — better endurance, better blood sugar, even longer life in some studies. Three reasons not to bank on that:
- The doses are huge. For their body weight, mice often get far more than people take.
- Lab mice are nothing like us. They are short-lived and bred to be nearly identical. A small lifespan bump in a mouse says little about a varied human living 80 years.
- Perfect lab conditions. Effects that show up in a controlled cage often vanish in messy real life.
Promising in mice is the starting line, not the finish line.
Blood vs the tissues that actually age
Here is the part the marketing skips. Raising NAD+ in your bloodstream is not the same as raising it in your muscles, brain, or liver — the places where ageing happens. Some studies show blood NAD+ shooting up while muscle barely moves. So even if the whole theory were right, a big blood number would not guarantee the organs that matter are benefiting.
What about NAD+ IV drips?
Clinics sell NAD+ infusions for anywhere from $200 to $1,000 or more per session, pitched for “energy” and “anti-ageing”. The evidence here is even thinner than for the pills — just a handful of tiny studies, no proof of real benefit, and the drips can feel unpleasant. It is the priciest, least-proven corner of the whole space.
Doses and cost: what people actually take
Typical doses in studies are about 250–900 mg a day for NMN and 300–1,000 mg for NR. Expect to pay roughly $40–90 a month, ongoing — for benefits that still are not proven in humans. (Again, not medical advice. Check with your doctor first, especially if you have a health condition.)
Cheaper things with stronger evidence
Want to support your NAD+ without the price tag? Exercise naturally raises NAD+ and the enzymes around it in your muscles — and it has far more proof behind it for healthy ageing than any powder. Plain niacin (vitamin B3) is a cheap precursor too. None of this is exciting marketing, but it is where the real evidence sits.
Is it safe?
Short-term, it looks reasonable. Studies running up to about a year report no serious problems, with only mild, occasional side effects like nausea or an upset stomach. The honest caveats:
- Long-term is untested. Most trials last weeks to months. Taking these for years has not been studied.
- Quality varies a lot. Pick a brand with third-party testing.
- One open question: because NAD+ helps cells grow, some researchers want more long-term safety data around cancer. That is unresolved — not proven harm, but worth knowing.
NMN’s rollercoaster with the FDA
This one is a genuine saga, and it tells you how unsettled the field is. In 2022, the FDA said NMN could not be sold as a supplement — because a drug company (Metro Biotech) had registered it for study as a drug, which legally blocks it from also being a supplement. Then in 2025, the FDA reversed course and said NMN is allowed as a supplement after all. So it is legal again today — but a molecule getting yanked and reinstated within three years is not the sign of a settled, well-understood product. (NR never had this problem.)
NMN vs NR: which one?
Both are NAD+ building blocks. Both raise blood NAD+. NR has a little more human research behind it; NMN has louder marketing and is, in theory, one step closer to NAD+. But there is no strong human evidence that either beats the other for any real health outcome. If you do try one, choose the brand with third-party purity testing over the one with the boldest promises.
The bottom line
NMN and NR reliably do one thing: raise the NAD+ level in your blood. Everything past that — living longer, ageing slower, more energy — is hope borrowed from mice, not proof from humans. If you want to try it, treat it as a low-confidence experiment: pick a third-party-tested brand, spend in proportion to the (thin) evidence, and never let a powder replace the basics. Training, sleep, and protein do far more for healthy ageing than any NAD+ pill on the market today.
